ABSTRACT

Basil (Ocimum basilicumL.) is a medicinal species used in several areas, such as food, medicines and cosmetics, and the understanding of its physiological behavior under environmental conditions is of paramount importance for the improvement of cultivation methods. The objective of this study was to evaluate the influence of different water availability under physiological, biochemical and metabolic characteristics, in three distinct genotypes: 'Alfavaca basilicão', 'Gennaro de menta' and 'Grecco à palla', during two different phenological stages (vegetative and reproductive). It was found that the water deficit promotes physiological changes to tolerate water stress, and the studied genotypes have different routes to achieve this physiological tolerance, which culminates in a distinct accumulation of metabolites in plants, and can be considered interesting if the final product is the production of essential oils.


La albahaca (Ocimum basilicum L.) es una planta medicinal utilizada en varias áreas: alimenticia, medicinal e industria cosmética; es de suma importancia el entendimiento de su comportamiento fisiológico bajo diferentes condiciones ambientales con el fin de mejorar los procesos del cultivo. El objetivo de este estudio fue evaluar la influencia de diferentes disponibilidades hídricas en las características fisiológicas, bioquímicas y metabólicas en tres genotipos de albahaca: "Alfavaca basilicão", "Gennaro de menta" y "Grecco à palla" durante dos etapas fenológicas (vegetativa y reproductiva). Fue encontrado que el déficit hídrico promueve cambios fisiológicos con el fin de tolerar el estrés hídrico. Los genotipos estudiados presentaron diferentes rutas para alcanzar esta tolerancia fisiológica, la cual culmina con distintas acumulaciones de metabolitos en las plantas, y puede ser considerado interesante si el producto final es la producción de aceites esenciales.

ABSTRACT

ABSTRACT Varronia curassavica, a subshrubby medicinal species associated with restinga in the Atlantic Forest, has been exploited by local people and the pharmaceutical industry. Indeed, restingas have experienced a continuous process of degradation, and thus, with species and ecosystem both at risk, efforts to support conservation actions are required. The present study aimed to evaluate aspects of V. curassavica reproductive biology. To accomplish this, morphological characterization was performed by monitoring flowering events. The availability of nectar and pollen, as well as the frequency and behavior of floral visitors and dispersers, was also evaluated. This species exhibits both heterostyly and protogyny. Anthesis is diurnal, and flowers last less than a day. The high number of flower and fruit abortions suggests that mechanisms, such as self-incompatibility intra-morphs and easily detached flowers, contribute to reduced fruit production. The high diversity of floral visitors indicate a generalist pollination syndrome. Diptera, Hymenoptera and Lepidoptera were the main pollinators, and nectar was the main resource sought by these insects. Fruits were dispersed by birds and ants. It can be concluded that the interaction of V. curassavica with several species is a key factor in its own survival and for maintaining the biological diversity of restinga.

Abstract:Phyllonoma laticuspis leaves are used in Carrizal de Bravo, Guerrero, Mexico, to heal skin lesions such as injuries and smallpox sequelae and to treat diabetes mellitus type 2, and organic extracts of these leaves have been reported to exert antibacterial effects. High demand of P. laticuspis as a medicinal plant has decreased its natural populations, and propagation of the species has not yet been reported. Therefore, the purpose of this study was to assess the vegetative propagation of the species through cutting and air layering, as well as its sexual propagation in a preserved population. For this, concentrations of 1 000, 4 000 and 6 000 ppm of a commercial root enhancer, with indole butyric acid (IBA), and a control treatment without IBA, were applied to the cuttings and air layers. Germination was evaluated under light and dark conditions using lots of freshly collected seeds and lots of seeds that had been stored for three months at 4 °C or 24 ± 2 °C. All experiments were performed in a completely randomized design. The cuttings did not develop roots in any concentration, whereas 100 % of the air layers rooted, displaying vigorous roots in the presence of 4 000 ppm IBA, after four month of treatment application. Regarding germination, more than 60 % of the freshly collected seeds germinated, whereas less than 20 % of the seeds stored at 4 °C, and close to 50 % of the seeds stored at 24 ± 2 °C germinated under light and dark conditions. No significant differences were observed between light and dark conditions, so they were categorized as indifferent photoblastic seeds. The observed moisture content of 13.5 % and germination behaviour as the response to cold storage, suggest that the resultant seed quality was intermediate. P. laticuspis propagation for short-term foliage production can be carried out in air layers, in populations with a high density of adult plants as a source of plant material and for the restoration of disturbed areas, in the same locality. On the other hand, large-scale seedling production, medium-term foliage production and preservation of species variability can be achieved using seeds. Rev. Biol. Trop. 65 (1): 9-19. ResumenLas hojas de Phyllonoma laticuspis se utilizan en Carrizal de Bravo, Gro. México, para curar lesiones de la piel como heridas, secuelas de viruela y afecciones de la diabetes mellitus tipo 2. Los extractos orgánicos de hojas, mostraron efectos antibacterianos. La gran demanda de P. laticuspis como planta medicinal, ha disminuido sus poblaciones naturales, y no existen reportes de la propagación de la especie. Por lo tanto, el propósito del presente trabajo fue evaluar la propagación vegetativa de la especie a través de estacas y acodos, y su propagación sexual en una población conservada. En la propagación por estacas y acodos se aplicaron concentraciones de 1 000, 4 000 y 6 000 ppm de un enraizador comercial con ácido indolbutírico (AIB), y un tratamiento control sin AIB. La germinación se evaluó en luz y oscuridad mediante el uso de lotes de semillas recién recolectadas y lotes de semillas almacenadas por tres meses a 4 °C y a 24 ± 2 ºC. Todos los experimentos se realizaron en un diseño completamente al azar. Las estacas no desarrollaron raíces en ninguna de las concentraciones, mientras que el 100 % de los acodos enraizaron, produciendo raíces vigorosas con la concentración de 4 000 ppm de AIB, a los cuatro meses de la aplicación. Con respecto a la germinación bajo condiciones de luz y oscuridad, las semillas recién recolectadas germinaron más del 60 %, mientras que las semillas almacenadas a 4 ºC menos del 20 % y las almacenadas a 24 ± 2 ºC cerca del 50 %. No hubieron diferencias significativas entre la germinación en luz y oscuridad, por lo que se consideraron como semillas fotoblásticas indiferentes. El contenido de humedad de 13.5 % y el comportamiento de la germinación en respuesta al almacenamiento en frío, sugieren la cualidad de semillas de tipo intermedias. La propagación de P. laticuspis para la producción de follaje a corto tiempo, puede hacerse por acodos en poblaciones con alta densidad de plantas adultas como fuente de material vegetal y para la restauración de áreas alteradas en la misma localidad. Mientras que la producción de follaje a mediano plazo y la conservación de la variabilidad de la especie, se puede lograr con la obtención masiva de plántulas a través de semillas.

Different explants of fenugreek, T. foenum-graecum L. (Var. RMt-303), were compared for their callus induction and subsequent shoot regeneration capabilities on Murashige and Skoog media supplemented with different phytohormones in varying concentration. The highest percentage of callus induction frequency was observed in 1ppm benzylaminopurine (BAP). Maximum shoots were induced on media supplemented with 0.5ppm BAP using leaf and stem tissues as explants. However, root tissues showed only callusing with no subsequent shooting. Cotyledonary node responded better than hypocotyls in terms of shoot induction on media supplemented with thidiazuron (0.1ppm). The callus was subjected to drought stress as simulated by reduced water potential of growth media due to addition of mannitol. Calli could withstand -2 MPa water potential till 30 days indicating that the drought stress tolerance mechanisms are functional in this variety. Chlorophyll a and b and total chlorophyll, proline and total phenolic contents, total peroxidase and catalase activities increased under stress conditions suggesting the tolerance of callus to drought stress. However, ascorbate peroxidase, guaiacol peroxidase activities were found to decrease slightly. Malondialdehyde and H2O2 contents were found to decrease while only a slight disturbance was found in membrane stability index. These results underline the mechanisms that are crucial for drought stress tolerance in fenugreek.

ABSTRACT

Various parameters including explant-type, medium compositions, use of phytohormones and additives were optimized for direct and indirect regeneration of E. ochreata, a medicinal orchid under threat. Protocorm-like-bodies (PLBs) proved to be the best explants for shoot initiation, proliferation and callus induction. Murashige and Skoog’s (MS) medium containing 2.5 mg L-1 6-benzylaminopurine (BAP), 1.0 mg L-1 kinetin (Kin) and additives (adenine sulfate, arginine, citric acid, 30 mg L-1 each and 50 mg L-1 ascorbic acid) was optimal for shoot multiplication (12.1 shoots and 7.1 PLBs per explant with synchronized growth), which also produced callus. Shoot number was further increased with three successive subcultures on same media and ~40 shoots per explant were achieved after 3 cycles of 30 days each. Additives and casein hydrolysate (CH) showed advantageous effects on indirect shoot regeneration via protocorm-derived callus. Optimum indirect regeneration was achieved on MS containing additives, 500 mg L-1 CH, 2.5 mg L-1 BAP and 1.0 mg L-1 Kin with 30 PLBs and 6 shoots per callus mass (~5 mm size). The shoots were rooted (70% frequency) on one by fourth-MS medium containing 2.0 mg L-1 indole-3-butyric acid, 200 mg L-1 activated charcoal and additives. The rooted plantlets were hardened and transferred to greenhouse with 63% survival rate. Flow-cytometry based DNA content analysis revealed that the ploidy levels were maintained in in vitro regenerated plants. This is the first report for in vitro plant regeneration in E. ochreata.

ABSTRACT

Vitex trifolia is a shrub species with popular use as a medicinal plant, for which leaves, roots and flowers have been reported to heal different distresses. The increasing exploitation of these plants has endangered its conservation, and has importantly justified the use of biotechnological tools for their propagation. Our aim was to present an efficient protocol for plant regeneration through organogenesis; and simultaneously, to analyze the genetic homogeneity of the established clonal lines by Randomly Amplified Polymorphic DNA (RAPD) and Inter Simple Sequence Repeat (ISSR) markers. Plantlet regeneration was achieved in callus cultures derived from stem, leaf and petiole explants of V. trifolia on a differently supple mented Murashige & Skoog medium, and incubated at 25±2ºC under a light intensity of 61µmol/m2s from cool white fluorescent lamps and a 16h photoperiod. The rate of shoot bud regeneration was positively correlated with the concentration of hormones in the nutrient media. Shoot buds regenerated more rapidly from stem and petiole explants as compared to leaf explants on medium containing 11.10µM BAP in combination with 0.54µMNAA. Addition of 135.74-271.50µM adenine sulphate (Ads) and 0.72-1.44µM gibberellic acid (GA3) to the culture medium increased the growth of shoot buds. The highest rate of shoot bud regeneration responses was obtained in stem explants using 11.10µM BAP in combination with 0.54µM NAA, 271.50µM Ads and 1.44µM GA3. In vitro rooting of the differentiated shoots was achieved in media containing 1.23µM indole butyric acid (IBA) with 2% (w/v) sucrose. Regenerated plantlets were successfully established in soil with 86% survival under field condition. Randomly Amplified Polymorphic DNA and Inter Simple Sequence Repeat markers analyses have confirmed the genetic uniformity of the regenerated plantlets derived from the second up to fifth subcultures. This protocol may help in mass propagation and conservation of this important medicinal plant of great therapeutic potential.


Vitex trifolia es una especie arbustiva de uso popular como planta medicinal, sus hojas, raíces y flores se han reportado para la cura de diferentes aflicciones. El aumento de la explotación de estas plantas ha puesto en peligro su conservación y ha justificado el uso de herramientas biotecnológicas para su propagación. El objetivo de esta investigación fue presentar un protocolo eficiente para la regeneración de estas plantas a través de la organogénesis, y analizar la homogeneidad genética de las líneas clonales establecidas por ADN polimórfico amplificado aleatoriamente (RAPD) mediante la repetición de marcadores de inter secuencia simple (ISSR). La regeneración de plántulas se logró en cultivos de callos derivados de explantes de tallo, hoja y pecíolo de V. trifolia en un medio diferenciado Murashige & Skoog, que se incubaron a 25±2ºC bajo una intensidad de luz de 61μmol/m2s con lámparas fluorescentes blancas y un fotoperíodo de 16h. La tasa de regeneración de brotes se correlacionó positivamente con la concentración de las hormonas en el medio nutritivo. Los brotes se regeneraron más rápidamente a partir de explantes de tallo y pecíolos en comparación con explantes de hoja. La mayor tasa de regeneración de brotes se obtuvo en los explantes de tallo utilizando 11.10μM BAP en combinación con 0.54μM NAA, 271.50μM Ads y 1.44μM GA3. Este protocolo puede ayudar a la propagación masiva y conservación de esta importante planta medicinal de gran potencial terapéutico.

Chlorophytum arundinaceum is an important medicinal plant and its tuberous roots are used for various health ailment treatments. It has become an endangered species in the Eastern Ghats, and a rare medicinal herb in India, due to its excessive collection from its natural habitat and its destructive harvesting techniques, coupled with poor seed germination and low vegetative multiplication ratio. In order to contribute to its production systems, an efficient protocol was developed for in vitro clonal propagation through shoot bud culture. For this, multiple shoots were induced from shoot bud explants on Murashige and Skoog’s medium supplemented with 2.5-3.0mg/L BAP, 0.01-0.1mg/L NAA and 3% (w/v) sucrose. Inclusion of Adenine Sulphate (25mg/L) in the culture medium improved the frequency of multiple shoot production and recovered the chlorotic symptoms of the leaves. Media having pH 5.9 and 4% sucrose showed significant improvement on shoot bud multiplication and growth. In vitro flowering was observed when the subcultures were carried out for over four months in the same multiplication media. Rooting was readily achieved upon transferring the shoots on to half- strength MS medium supplemented with 0.1mg/L IBA and 2% (w/v) sucrose. Micropropagated plantlets were hardened in the green house, successfully established, and flowered in the field. Chlorophytum arundinaceum es una planta medicinal importante y sus raíces se utilizan en diversos tratamientos contra enfermedades. Se ha convertido en una especie en peligro de extinción en el Ghats Oriental y una hierba medicinal rara en la India, debido a la recolecta excesiva en su hábitat natural y la manera destructiva de cosecharla, asociado con una mala germinación y pobre multiplicación vegetativa. Para contribuir con sus sistemas de producción, se desarrolló un protocolo eficiente para la propagación clonal in vitro a través del cultivo de brotes. Para ello, los retoños múltiples fueron inducidos a partir de sus brotes en un medio Murashige y Skoog enriquecido con 2.5-3.0mg/L de BAP, 0.01-0.1mg/L de NAA y el 3% (w/v) sucrosa. La inclusión de sulfato de adenina (25mg/L) en el medio de cultivo mejoró la frecuencia de producción de brotes múltiples y se recuperaron los síntomas de clorosis de las hojas. Los medios con un pH de 5.9 y 4% de sucrosa mostraron una mejoría significativa en la multiplicación y crecimiento de las yemas. En la floración in vitro se observó cuando los subcultivos se llevaron a cabo durante más de cuatro meses para los mismos medios de multiplicación. El enraizamiento se logró fácilmente al transferir los brotes a un medio MS de intensidad media enriquecido con 0.1 mg/l de IBA y 2% (w/v) de sucrosa. Las plántulas micropropagadas maduraron en el invernadero, se establecieron exitosamente y florearon en el campo. Este método se podría aplicar para la conservación y propagación clonal con el fin de satisfacer la demanda de material de siembra.

ABSTRACT

A erva-cidreira [Lippia alba (Mill.) N.E.Brown] é uma espécie nativa utilizada como planta medicinal em todo o Brasil. O citral é responsável pela ação sedativa da erva cidreira, sendo um dos principais componentes do óleo essencial. O objetivo deste trabalho foi avaliar o comportamento agronômico, teor e rendimento de óleo essencial e de citral em 17 acessos de L. alba da coleção da Universidade de Brasília - UnB. O experimento foi realizado no período de 19/02/2005 a 18/03/2006, em latossolo vermelho, no Distrito Federal. O delineamento experimental utilizado foi o de blocos ao acaso, com três repetições, contendo três plantas por parcela. Foram avaliados os parâmetros área foliar, comprimento da haste, produção de massa fresca (folhas e hastes), produção de massa foliar seca, teor de óleo essencial e de citral, rendimento estimado de óleo essencial e de citral por planta, além da caracterização do perfil aromático do óleo essencial. Os acessos L.41 (0,75 por cento), L.45 (0,66 por cento) e L.08 (0,62 por cento) apresentaram os maiores teores de óleo essencial. O acesso L.45 (1,32 g pl-1) apresentou o maior rendimento de óleo, seguido dos acessos L.47 (0,73 g pl-1), L.41 (0,67 g pl-1), L.34 (0,67 g pl-1) e L.33 (0,62 g pl-1). Os acessos apresentaram teores de citral variando de 51,7 por cento (L.34) a 75,1 por cento (L.09), com média de 63,6 por cento e predominância do isômero geranial (36,4 por cento) sobre o neral (27,8 por cento). O acesso L.45 (0,83 g pl-1) apresentou o maior rendimento de citral. Treze acessos apresentaram o quimiotipo citral; três o quimiotipo citral-limoneno e apenas um apresentou o quimiotipo citral-mirceno. Os resultados evidenciam o potencial de produção de citral do acesso L.45, o qual poderá ser utilizado para melhorar a qualidade da matéria prima de erva-cidreira utilizada nos programas de fitoterapia no país ou em programas de melhoramento genético para esta finalidade.


"Erva-cidreira" [Lippia alba (Mill.) N.E.Brown] is a native species used as a medicinal plant all over Brazil. Citral, a major constituent of L. alba essential oil, is responsible for its sedative activity. The aim of this study was to evaluate the agronomic behavior, content and yield of essential oil and citral in 17 accessions of L. alba from the germplasm bank of University of Brasília - UnB, Brazil. The experiment was carried out from 02/19/2005 to 03/18/2006, in red latosol, at the Federal District, Brazil. The adopted experimental design was in randomized blocks, with three replicates and three plants per plot. The following parameters were evaluated: leaf area, stem length, fresh mass (leaves and stems) production, dry leaf mass production, essential oil and citral content, estimated yield of essential oil and citral per plant, besides characterization of the essential oil aromatic profile. Accessions L41 (0.75 percent), L45 (0.66 percent) and L08 (0.62 percent) showed the highest essential oil content. Accessions L45 (1.32 g pl-1) presented the highest essential oil yield, followed by L47 (0.73 g pl-1), L41 (0.67 g pl-1), L34 (0.67 g pl-1) and L33 (0.62 g pl-1). Citral content varied from 51.7 percent (L34) to 75.1 percent (L09), with an average of 63.6 percent and predominance of the isomer geranial (36.4 percent) over neral (27.8 percent). Accession L45 (0.83 g pl-1) showed the highest citral yield. Thirteen accessions presented the chemotype citral; three, the chemotype citral-limonene, and only one had the chemotype citral-myrcene. The results evidenced the potential of accession L45 to produce citral, which can be used to improve the quality of "erva-cidreira" raw material in phytotherapy programs in the country or in breeding programs.

ABSTRACT

A caracterização morfoanatômica e a correta identificação das plantas medicinais são fundamentais para o controle de qualidade dos fitoterápicos. No presente trabalho é caracterizada a anatomia do lenho visando à identificação de seis espécies arbóreas com potencial medicinal. As amostras do lenho foram extraídas por método não destrutivo do tronco das árvores das espécies desenvolvidas em áreas de preservação. Em laboratório as amostras do lenho foram processadas para análises microscópicas do lenho, aplicando as metodologias para estudos de anatomia. Os resultados mostraram que o parênquima axial foi o parâmetro anatômico mais efetivo para a distinção das espécies através da análise do seu lenho. No trabalho discute-se a importância da aplicação da anatomia do lenho como ferramenta na identificação de espécies arbóreas medicinais.


The morphoanatomical characterization and the correct identification of medicinal plants are fundamental for the quality control of phytotherapics. In the present work, wood anatomy was characterized in order to identify six tree species with medicinal potential. Wood samples were collected through a non-destructive method from the trunk of trees developed in conservation areas. In the laboratory, wood samples were processed for microscopic analysis by applying the methods for anatomical studies. Axial parenchyma was the most effective anatomical parameter to distinguish species through wood analysis. In this work, the importance of wood anatomy as a tool for the identification of medicinal tree species was discussed.

ABSTRACT

Folhas de Myrcia multiflora (Lam.) DC. são usadas na medicina popular como hipoglicemiantes. O objetivo deste trabalho foi caracterizar morfológica e anatomicamente as folhas desta planta, de modo que os dados obtidos possam ser utilizados como referência em exames de controle de qualidade de amostras de fármacos, com vistas a verificar a autenticidade. Folhas inteiras foram diafanizadas e coradas para o estudo da nervação. Secções transversais do pecíolo e transversais e paradérmicas da lâmina foliar foram analisadas em microscópio óptico (MO) e a superfície do limbo foi observada, também, em microscopia eletrônica de varredura (MEV). Foram aplicados testes histoquímicos em material fresco, para identificação e localização de glicídios, amido, taninos, lignina, cristais e sílica. Morfologicamente, a folha é simples, oval-elíptica, com margem inteira, base aguda, ápice acuminado e textura cartácea. A venação é do tipo camptódromo-broquidódromo. Anatomicamente, a folha é hipostomática, com mesofilo compacto e dorsiventral, com três estratos de parênquima paliçádico. A epiderme é uniestratificada, silicificada em algumas regiões e as células exibem paredes anticlinais retas. Em posição subepidérmica ocorrem numerosas cavidades secretoras de óleos essenciais. Os feixes vasculares são colaterais e acompanhados por séries cristalíferas. Os dados obtidos são comparados com os de outras espécies de Myrtaceae e conclui-se que as características morfológicas e anatômicas de M. multiflora contribuem para a diagnose.


Myrcia multiflora (Lam.) DC. leaves have been used in folk medicine as hypoglycemic. The aim of this work is to describe morphoanatomically the leaves of this plant in order to use the obtained data as reference in quality control tests of drug samples, investigating their authenticity. Whole leaves were diaphanized and stained for venation study. Petiole transverse sections and leaf blade transverse and paradermal sections were analyzed under an optical microscope (OM). Leaf surface was also observed by scanning electron microscopy (SEM). Histochemical tests were done using fresh material to identify and locate carbohydrates, starch, tannins, lignin, crystals and silica. Morphologically, the leaf is simple, oval-elliptical, with entire margin, acute base, acuminate apex and chartaceous texture. The venation pattern is camptodromous-brochidodromous. Anatomically, the leaf is hipostomatic, with compact and dorsiventral mesophyll, containing three layers of palisade parenchyma. The epidermis is uniseriate, silicified in some regions and presents cells with straight anticlinal walls. In the subepidermal position, there are several essential oil secretory cavities. Vascular bundles are collateral and surrounded by crystalliferous series. The obtained data were compared to those of other Myrtaceae species , leading to the conclusion that, the morphoanatomical traits of M. multiflora contribute to its diagnosis.

ABSTRACT

Este trabalho teve como objetivo realizar levantamento das plantas medicinais utilizadas pela comunidade de São José de Espinharas, a fim de registrar e preservar o conhecimento popular. A metodologia usada foi de entrevistas semi-estruturadas, com observações participantes, coleta e identificação do material botânico e preparação de exsicatas, resultando em lista de 82 espécies de 38 famílias diferentes. São fornecidas as informações nomenclatura popular e botânica, uso terapêutico, parte utilizada, forma de uso, modo de preparo e doenças tratadas. As raízes (30 por cento) foram ás partes mais utilizadas e a forma de preparo foi lambedor (32 por cento). O conhecimento sobre os usos e modos de preparo provém em geral, dos familiares (85 por cento). Com esses resultados, verifica-se a interação da população local com a flora e utilização relacionada a aspectos sociais, econômicos, culturais e às mudanças ambientais.


The aim of this study was to perform a survey of medicinal plants used by the community of São José de Espinharas, Paraíba State, Brazil, in order to record and preserve the folk knowledge. The adopted methodology was based on semi-structured interviews, with participating observations, botanical material collection and identification, and voucher preparation, resulting in a list of 82 species of 38 different families. The following information was provided: folk and botanical nomenclature, therapeutic application, used plant part, forms of use, method of preparation and treated diseases. Roots (30 percent) constituted the part most frequently used and syrup (32 percent) was the predominant method of preparation. The knowledge of uses and methods of preparation are generally handed down in the family (85 percent). Based on those results, there is an interaction of the local population with the flora and its use is related to social, economical and cultural aspects and environmental changes.

ABSTRACT

As plantas são fontes importantes de produtos naturais biologicamente ativos. Dentre as plantas usadas na medicina popular a Anonna squamosa conhecida como fruta-do-conde é citada como tendo várias ações medicinais, dentre elas a atividade inseticida e anti-helmíntica. Dentro desta perspectiva, objetivou-se determinar a atividade anti-helmíntica dos extratos aquosos (EA) e etanólicos (EE) das folhas da fruta-do-conde sobre o nematóide de aves Ascaridia galli, in vitro e in vivo. No primeiro, os nematóides foram colocados em placa de Petri contendo diferentes concentrações dos extratos e no segundo foram utilizadas seis galinhas poedeiras por grupo, as quais foram administrados10 mL Kg-1 dos extratos. No teste in vitro o EA da A. squamosa nas concentrações 2,4 e 9,6 mg mL-1 foi capaz de matar 63,33 por cento e 53,33 por cento dos nematóides, respectivamente. O EE não produziu efeito significativo. No teste in vivo, o percentual de eliminação do EA foi de 39 por cento e do EE de 20 por cento. Estes dados sugerem que neste caso a substância responsável pela mortalidade dos parasitos esteja em maior concentração na fração aquosa. Desta maneira, acredita-se que o EA de A. squamosa apresenta uma atividade anti-helmíntica potencial sobre o A. galli.


Plants are important sources of biologically active natural products. Among the plants used in popular medicine, Annona squamosa, known as sweetsop, is reported to have several medicinal actions such as insecticidal and anthelmintic activity. Therefore, this study aimed to assess the anthelmintic activity of aqueous (AE) and ethanolic (EE) extracts from sweetsop leaves on the chicken roundworm Ascaridia galli, both in vitro and in vivo. In the former, nematodes were placed on a Petri plate containing different concentrations of the extracts; in the in vivo test, six egg-laying chickens per group received 10 mL Kg-1 of the extracts. In vitro results indicated that A. squamosa AE at the concentrations 2.4 and 9.6 mg mL-1 could kill 63.33 percent and 53.33 percent nematodes, respectively. However, EE did not have any significant effect. According to the in vivo test, the elimination percentage for AE was 39 percent and for EE, 20 percent. These data suggest that the substance responsible for parasite mortality was present at a higher concentration in the aqueous fraction. Thus, A. squamosa AE is believed to have a potential anthelmintic activity on A. galli.

El propósito del presente trabajo fue determinar la potencia antimicrobiana de extractos alcohólicos de plantas utilizadas popularmente en Argentina como antisépticos y antiinflamatorios: Dasyphyllum diaconthoides, Erythrina cristagalli, Larrea cuneifolia, Larrea divaricata, Phytolacca dioica, Pithecoctenium cynanchoides, Prosopanche americana, Schinus molle, Schkuhria pinnata, Senna aphylla y Solidago chilensis. La inhibición del crecimiento bacteriano se determinó a través de ensayos de difusión en agar, macrodilución en medio sólido y microdilución en medio líquido frente a 47 aislamientos clínicos multirresistentes a antibióticos, obtenidos de pacientes de un hospital de Tucumán, Argentina: Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis, Enterobacter cloacae, Serratia marcescens, Morganella morganii, Acinetobacter baumannii, Pseudomonas aeruginosa, Stenotrophomonas maltophilia. De acuerdo con los valores de concentración inhibitoria mínima (CIM), tres de las once especies ensayadas fueron las más activas: L. divaricata, L. cuneifolia, y S. aphylla (CIM de 25 a 200 µg/mL). P. mirabilis, A. baumanni y S. maltophilia fueron las cepas más susceptibles con valores de CIM entre 25 y 50 µg/mL seguido por P. aeruginosa con valores de CIM de 50 a 100 µg/mL. Los valores de concentración bactericida mínima (CBM) fueron semejantes o dos veces superiores a los valores de CIM. Mediante ensayos bioautográficos se comprobó que los extractos más activos presentaban al menos dos principios antimicrobianos. Análisis fitoquímicos indican que estos compuestos son de naturaleza fenólica. Los resultados obtenidos justificarían el uso de estos extractos para el tratamiento de infecciones bacterianas, especialmente aquellas de origen dérmico.


The present study was conducted to investigate antimicrobial activity of alcoholic extracts of Argentine medicinal plant species (Dasyphyllum diacanthoides, Erythrina cristagalli, Larrea cuneifolia, Larrea divaricata, Phytolacca dioica, Pithecoctenium cynanchoides, Prosopanche americana, Schinus molle, Schkuhria pinnata, Senna aphylla and Solidago chilensis) against multidrug resistant human pathogen gram negative bacteria isolated from a Hospital in Tucumán, Argentina. Inhibition of bacterial growth was investigated using disc diffusion, agar macrodilution and broth microdilution methods against multiresistant clinical isolates of nine different specie of gram negative bacteria: Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis, Enterobacter cloacae, Serratia marcescens, Morganella morganii, Acinetobacter baumannii, Pseudomonas aeruginosa, Stenotrophomonas maltophilia. A significant antimicrobial activity was found in three of the eleven plant species studied. Based on minimal inhibitory concentration (MIC) values, three plant species, L. divaricata, L. cuneifolia, and S. aphylla were the most potent ones with MIC values between 25-200 µg/mL. Overall, P. mirabilis, M. morganii and P. aeruginosa isolates were the most susceptible to these extracts with MIC values of 25 to 50 µg/mL. All extracts showed significant inhibitory activities on bacteria growth in a dose phenolic compound-dependent fashion. The minimal bactericidal concentration (MBC) values were identical to the MIC values or twofold higher than the corresponding MIC. Contact bioautography indicated that crude extracts possess several major antibacterial components. Phytochemical screening showed that the bioactive compounds correspond to polyphenols. Investigations are in progress to purify the bioactive principles.

ABSTRACT

Se evaluó cuantitativamente el uso de plantas medicinales con el propósito de documentar la importancia relativa de dichas especies y estimar el estado actual del conocimiento tradicional de las comunidades campesinas de la vereda San Isidro, municipio de San José de Pare, Boyacá. Esta zona en particular, se considera un sector agrícola de alta confluencia y denota una apropiación del conocimiento de sus pobladores en el uso y manejo de las especies vegetales de la región. Se aplicaron aleatoriamente 180 encuestas a personas entre los 20 y 80 años, en donde se investigó respecto al uso y manejo de las plantas. Se aplicaron entrevistas programadas a aquellas personas que manejan en gran parte la demanda de plantas medicinales en la región. Los datos obtenidos a partir del consenso de informantes, fueron analizados mediante el índice de valor de uso (IVU) y el nivel de uso significativo de TRAMIL. Se registraron 84 especies con 78 géneros, agrupadas en 42 familias botánicas entre medicinales, aromáticas y mágicoreligiosas; las familias mejor representadas fueron Lamiaceae, Asteraceae, Apiaceae, Solanaceae y Rutaceae. Se reportaron 10 especies con un mayor nivel de uso significativo y 25 especies con un mayor IVU. La documentación de los usos de las plantas medicinales en el área de influencia, revela que el conocimiento tradicional continúa profundamente arraigado entre la comunidad y se mantiene el saber popular a manos de curanderos y madres cabeza de familia.

ABSTRACT

A protocol was developed for high frequency plant regeneration in H. patulum by shoot-tip culture. H. patulum plants were collected from a wild source growing at high altitude in the eastern Himalayas. Multiple buds were initiated from shoot-tips cultured on Murashige and Skoog's medium supplemented with BAP, kinetin. Addition of thiamin HCI, Ca-pantothenate and biotin enhanced multiple shoot formation. Upon transfer to phytohormone free liquid medium following a brief exposure to auxin, root formation occurred from the micro shoots . Rooted plants were hardened and transferred to soil. Regeneration potentiality was found to be constant throughout the year in long term cultures.

ABSTRACT

Rhamnose, xylose, arabinose, glucose, fructose, galactose, sucrose, and rhafinose as well as traces of cellobiose and galacturonic acid were identified [TLC] in 90% ethanol extract of Chenopodium murale, Cyperus alopecuroides, Desmostachya bipinnata and Tamarix nilotica. Water soluble polysaccharides were prepared by hot extraction of the 4 plants, the total extractable pectins were determined, and component sugars detected [TLC] after hydrolysis were found to be identical in the 4 plants and similar to those of the corresponding free sugar extractives except for the absence of sucrose, rhafinose, cellobiose and fructose. Crude protein percentage were determined in the 4 plants. Amino acids contents of the 4 plants were almost similar, and glutamic acid was found to be the major component. Lipoidal composition of the 4 plants was studied, fatty acids identified by GLC of the methylated saponifiable fractions revealed qualitative and quantitative variations between the 4 plants, but palmitic acid was the major component in all of the investigated plants. Alpha-amyrin and beta-sitosterol were isolated from the unsaponifiable fraction of C. alopecuroids

ABSTRACT

Glycyrrhizin, a triterpenoid glycoside and Licorice from Glycyrrhiza glabra and Ammonium salt of Glycyrrhizic acid (Sigma) were tested for antiviral activity on three strains of Japanese encephalitis virus (JEV), Nakayama, P-20778 and 821564 XY48. Purified glycyrrhizin (M.w. 822.92) inhibited plaque formation in all the three strains of JEV at a concentration of 500 micrograms/ml at 96 hrs, Similar effect was observed at 1000 micrograms/ml concentration with Licorice and Ammonium salt of glycyrrhizic acid. The minimal inhibitory concentrations were not toxic to porcine stable kidney (PS) and human cervical carcinoma (HeLa) cell lines. Cyctotoxicity of these chemicals was evaluated by trypan blue dye exclusion test which indicated subtoxic concentrations at 5,000 micrograms/ml at 96 hrs and toxic concentrations were 10,000 micrograms/ml at the same time period for the host cells PS. Thus the indigenously purified glycyrrhizin seems to be more potent antiviral agent than Licorice and ammonium salt of glycyrrhizic acid (Sigma) for JEV 'in vitro'.

ABSTRACT

O Stryphnodendron basbatiman (SB) é uma árvore geralmente encontrada e utilizada no cerrado mineiro, cuja casca supõe-se ter propriedades cicatrizantes. O objetivo do presente trabalho é estudar a ação do S. basbatiman em ratos. Foram utilizados 8 ratos albinos (Rattus norvegicus), variedade Wistar, sendo 4 para o grupo controle e 4 para o grupo tratado com o estrato da planta. Os animais foram sacrificados 3 e 7 dias de pós operatórios. As peças foram tratadas, coradas em hematoxilina e eosina, sendo, então, analisadas histopatologicamente. No grupo tratado as cicatrizações foram mais rápidas. Esses resultados sugerem que o uso do extrato de SB pode ser util no processo de cicatrização de feridas.

ABSTRACT

A double masked placebo controlled multicentric randomised clinical trial was conducted in 157 patients suffering from different eye ailments namely, cataract, dry eye syndrome, ocular asthenia, refractive errors and allergic conjunctivitis with a herbal eye drop preparation and placebo. In both dry eye syndrome and ocular asthenia the herbal eye drop preparation was found better than placebo. In a few cases of cataract, the vision seems to be improved. In early myopia it seems to correct the refractive errors while in high myopic conditions it controlled the progressive deteriorations. Subjective improvements were also noted with hypermetropia, presbyopia and astigmatism. Its healing capacity in allergic conjunctivitis was certainly better than placebo. It had no short term or long term side-effects. It is considered to be a useful drug in all conditions studied.
